The current novel molecular DNA cloning procedure – during which a given DNA fragment (DNA stretch subjected to cloning, also called “insert”) aimed to be multiplied is introduced into a vector DNA (“vehicle”) without using in vitro added enzymes such as restriction endonucleases, ligases, DNA polymerases and 5′ exonucleases – is particularly suitable for generating novel DNA constructs in a relatively fast and cost-effective way, as it performs DNA cloning without using in vitro added enzymes. Hence, the solution is especially suitable for laboratories with limited resources. Another advantage of the solution is that it requires no specific recognition sites for restriction enzymes on the insert and vector DNA sequences, which is particularly problematic in case of cloning long DNA fragments.

Outline
Mycoplasma anserisalpingitidis is a pathogen lurking in geese which poses a threat primarily to the health of geese and, less commonly, ducks. Inflammation and atrophy develop in the cloaca and genitals of infected animals, the number of fertile eggs decreases, eggshell formation is impaired, and embryo mortality increases. Stress, which is an inevitable part of animal husbandry, especially large-scale but also barnyard animal husbandry, greatly increases the susceptibility of animals to both infection and the development of severe symptoms. In addition to congestion, inadequate housing conditions and the behavioral changes (mocking) associated with the mating season are also sources of stress in the life of our geese. M. anserisalpingitidis is likely to be ubiquitous in all of the major goose-breeding areas of the world: it has also been detected in Hungary, China, France and Vietnam. Until now, there has not been a sufficiently reliable method available against infection: despite proper housing and handling conditions, it can appear in any yard. Our solution is a vaccine to prevent the development of infection and / or symptomatic disease. Immunization of animals is easy and effective because the vaccine can be delivered into the cloaca or even sprayed onto the deep litter. The production of the active substance and formulation of the vaccine does not require special technology.

The invention further relates to the use of the novel strain PPV1 for eliciting an immune response. The invention further relates to the use of the new strain PPV1 for the preparation of vaccine compositions.

Members of the parvavirus family are small (18–26 nm in diameter), enveloped viruses with a linear single-stranded DNA genome composed of approximately 4–6 kilobases (kb.) in their capsid with icosahedral symmetry. PPV1, which can infect and cause disease in pigs, has long been known and occurs worldwide. Due to extensive and effective vaccinations, the pathogen has been reduced, but in recent years a variant of the virus, PPVla, has emerged which, due to a change in the amino acid sequence of the capsid protein, can cause reproductive defects through breakthrough vaccine protection. Due to unexpected detection, a new strain of the virus appeared in a Hungarian pig herd, which caused a significant health problem in the herd, mainly related to the vitality of the newborn piglets, and the known vaccines were not effective enough against it.

Outline
The field of extracellular vesicles (EVs) currently attracts substantial attention in biomedicine due to the proposed role of EVs in various biological processes and their potential of serving as biomarkers for diseases. However, the availability of approaches for reliable and reproducible standardised quantification of EVs is limited. Therefore, current interest in EV research urges reliable tools of standardization and accurate enumeration of EVs, preferably on the basis of lipid quantification. By definition, EVs are surrounded by phospholipid bilayers, therefore, lipids (such as phospholipids and cholesterol) are essential components of all EVs. The method of the invention avoids the overestimation of EV concentration based on the protein measurement, as it focuses on the defining component of EVs, the lipid bilayer. Our method can be used virtually in any standard laboratories where a fume hood, a thermoblock, and a spectrophotometer are available. The application does not require expensive equipment, therefore it can be an easy, reliable and quick method for quantification of EVs and standardisation of EV experiments.

During the mentioned marking phase, the mentioned nucleic acid sequences are sorted in groups, within the groups, compounds are formed by combining nucleic acid sequences, then a compound is selected from each group, and the selected compounds are formed into a marker compound. In a preferable case, the mentioned marker compound contains polymer, preferably resin, more preferably vegetable resin.
